そして従来は、上記支持体の針状体に細胞塊を突き刺すため、細胞塊を吸い込んだピペットや、ロボットアーム、ピンセットを用いて人手による作業が行われていた(特許文献1)。
このような問題に鑑み、本発明は自動的に細胞塊を支持体の針状体に突き刺して、細胞の立体構造体を得ることが可能な細胞の立体構造体製造装置を提供するものである。
上記吸引ノズルは、先端に1つの細胞塊を吸着する管状の吸着部を備えるとともに、該吸着部の内径を、上記細胞塊の外径より小径に、かつ上記支持体の針状体の外周径よりも大径とし、
上記制御手段は、上記収容プレートの収容凹部において上記吸引ノズルの吸着部に上記細胞塊を吸着保持させると、上記移動手段によって該吸引ノズルを上記支持体における所要の針状体の上方に移動させ、
さらに該吸引ノズルを針状体の軸方向に移動させて上記細胞塊に針状体を突き刺し、上記細胞塊を貫通した針状体を上記吸着部の内部に挿入させることを特徴としている。
その際、上記吸着ノズルの吸着部の内径を細胞塊の外径よりも小径に、かつ針状体の外周径よりも大径とし、上記移動手段は細胞塊を吸着保持した吸引ノズルを針状体の軸方向に移動させるため、細胞塊を確実に針状体に突き刺すことが可能となっている。
この立体構造体製造装置2では、後に詳しく述べる吸引ノズル19を用いて、図2に示す収容プレート3より1つずつ細胞塊1を取り出し、図3(a)に示す支持体4の針状体6に1つずつ細胞塊1を突き刺すことで、図3(b)に示すような細胞の立体構造体1Aを製造するものとなっている。
上記収容プレート3は、透明な部材によって製造され、縦横に複数の収容凹部3aが形成されている。図2(b)に示すように各収容凹部3aは底面が球面状の凹部によって構成され、各収容凹部3aの内部には培養液とともに略球状の細胞塊1が1つずつ収容されるようになっている。
また本実施例では、部分的に異なる種類の異種細胞塊1bを混在させて立体構造体1Aを製造することが可能であり、そのために、上記細胞塊1を収容する収容プレート3と、上記異種細胞塊1bを収容する異種収容プレート3bとを準備できるようになっている。
この支持体4は、上記立体構造体製造装置2において使用される際には図1に示す収容容器7に収容されるようになっており、この収容容器7は有底で上部が開口した容器となっており、上記支持体4を収容すると上記板状部材5が嵌合して位置決めされるようになっている。
上記針状体6は金属製で、それぞれに複数個(本実施例では4個)の細胞塊1を突き刺すことが可能な長さを有しており、また隣接する針状体6と針状体6との間隔は、突き刺された細胞塊1同士が相互に密着する程度の間隔、つまり細胞塊1の1個分程度の間隔に設定されている。
そして、各針状体6にそれぞれ複数個の細胞塊1を突き刺し、細胞塊1を針状体6の根元側から先端側へと積み重ねることで、図3(b)に示すような細胞の立体構造体1Aが得られるようになっている。このとき適宜上記異種細胞塊1bを組み合わせることで、異種細胞塊1bが混在する細胞の立体構造体1Aを得ることが可能となっている。
なお上記針状体6は、板状部材5に対してほぼ垂直に立設されているが、場合によっては図3(a)におけるAで示す針状体6のように、大きく傾いたものも含まれている。
このような構成を有する立体構造体製造装置2は、クリーンベンチ11に隣接して設けられた制御手段21によって制御され、この制御手段21はパーソナルコンピューター21aなどの端末によって各種の設定等が行われるようになっている。
上記クリーンベンチ11は図示しない開閉扉によって開閉可能となっており、上記供給マガジン12や回収マガジン13、上記支持体4を収容した上記収容容器7を搬出入することが可能となっている。
またクリーンベンチ11の上部には無菌エアを供給する図示しない無菌エア供給手段が設けられ、上記開閉扉が閉鎖された状態において、クリーンベンチ11の内部に上方から下方へと向かう無菌エアによる一方向流が形成されるようになっている。
上記第1、第3支持手段14、16は同じ高さに配置されるとともに、上記収容プレート3の4隅を下方から支持する4つの支持部材によって構成され、上記移送手段17はこの第1、第3支持手段14、16に対して上方から収容プレート3を載置するようになっている。
一方、上記第2支持手段15は第1、第3支持手段14、16よりも若干上方に設けられており、異種収容プレート3bの4隅を下方から支持する出没可能な係合爪15aを備えている。
上記第2支持手段15は、上記係合爪15aが突出することで異種収容プレート3bを下方から支持するようになっており、上記第2支持手段15から上記移送手段17へと異種収容プレート3bを受け渡す際や、移送手段17から第2支持手段15へと異種収容プレート3bを移載する際には、上記係合爪15aが退没して異種収容プレート3bの上下方向への移動を許容するようになっている。
上記昇降テーブル24は上記第1~第3支持手段14~16に収容プレート3や異種収容プレート3bを支持させた際は下降し、上記収容プレート3や異種収容プレート3bを移送する際には、第1、第3支持手段14、16より上方で第2支持手段15よりも下方に位置して、その状態で上記スライド機構22の作動により収容プレート3や異種収容プレート3bを移動させるようになっている。
上記載置台18には、上記支持体4を収容する収容容器7を位置決めして載置するようになっており、上記収容容器7を照明するリング照明25が、該リング照明25を旋回移動させる旋回手段26を介して設けられ、上記リング照明25を上記収容容器7の上方とここから退避した位置との間で旋回移動させるようになっている。
上記吸引ノズル19は、先端に1つの細胞塊を吸着する管状の吸着部19aと、上記移動手段20に保持される本体部19bを備え、固定手段20aにより上記移動手段20に交換可能に装着されている。
上記吸着部19aは、その内径を上記細胞塊1の外径より小径に、かつ上記支持体4の針状体6の外周径よりも大径としている。このような寸法とすることにより、図5(a)に示すように、先端に1つの細胞塊1を吸着させることができ、さらに、図5(b)に示すように、細胞塊1を貫通した針状体6を吸着部19aの内部に挿入させて、細胞塊1を押し込むことが可能となっている。
上記X-Yユニット34は、上記移送手段17の移送方向と直交する方向に可動支持部35を移動させるY方向機構34aと、該Y方向機構34aをその移動方向とは直交する方向へ移動させるX方向機構34bによって構成されている。
上記昇降手段36の可動部分に上記固定手段20aを介して吸引ノズル19が固定され、上記可動支持部35をX-Yユニット34により水平方向に移動させることで、上記吸引ノズル19を第3支持手段16に支持された収容プレート3または異種収容プレート3aと、載置台18に載置された収容容器7との間で移動させ、かつ、上記吸引ノズル19を昇降手段36により上下に昇降させることで、上記収容プレート3や異種収容プレート3bの収容凹部3a内の細胞塊1や異種細胞塊1bを吸着保持させたり、保持した細胞塊1や異種細胞塊1bを上記支持体4の針状体6に突き刺すことができるようになっている。
またパネル照明37は、スライド機構37aによって第3支持手段16の上方から移送手段17の移送方向側方に往復動するようになっており、第3支持手段16に支持された収容プレート3や異種収容プレート3bへの上記吸引ノズル19の下降を阻害しないように退避可能となっている。
上記下方カメラ38は、上記吸引ノズル19の真下に位置するように配置されており、該下方カメラ38と吸引ノズル19との間に上記第3支持手段16に支持された収容プレート3や異種収容プレート3bおよびパネル照明37が位置するようになっている。
上記下方カメラ38は、透明な収容プレート3や異種収容プレート3bを下方から撮影するようになっている。その撮影範囲は、図2(c)に示すように1つの収容凹部3aを撮影可能な範囲に設定されており、視野の中心は図2(c)に示す2本の中心線の交点となっている。
そして下方カメラ38が撮影した画像は上記制御手段21に送信され、制御手段21ではこの収容凹部3aに収容された細胞塊1の収容凹部3a内における位置を認識するとともに、該細胞塊1の外径およびその形状を認識するようになっている。
また、収容プレート3やパネル照明37が間に位置しない状態では、図6に示すように吸引ノズル19に装着された吸着部19aの先端を下方から撮影することが可能である。
そして上方カメラ39が撮影した画像は上記制御手段21に送信され、制御手段21ではこの支持体4に設けられた針状体6の先端位置を認識するようになっている。
まず、作業者は細胞塊1が収容された収容プレート3を収納した供給マガジン12と空の回収マガジン13をセットするとともに、上記第2支持手段15には異種細胞塊1bが収容された異種収容プレート3bを載置する。また作業者は支持体4を収容した収容容器7を載置台18上に位置決めし、上記吸引ノズル19には新たな吸着部19aを装着する。
また、事前に上記制御手段21に接続されたパーソナルコンピューター21aを用いて、製造する細胞の立体構造体1Aの構造の設定を行い、図3(b)に示すような細胞塊1と異種細胞塊1bとの配置を設定する。
制御手段21は下方カメラ38の視野中心に対して上記吸着部19aの中心がどれだけずれているかを画像処理によって測定し、そのずれ量を記憶する。このずれ量は、移動手段20のX-Yユニット34の作動により、吸引ノズル19を細胞塊1や異種細胞塊1bおよび針状体6の位置に位置決めする際に加味するようになっている。
次に、上記移送手段17の昇降テーブル24が昇降して、上記供給マガジン12から1枚の収容プレート3を取り出し、該収容プレート3をそのまま第3支持手段16へと移送する。
このとき、X-Yユニット34はあらかじめ記憶された収容プレート3の各収容凹部3aの位置に応じて下方カメラ38を移動させ、下方カメラ38の視野内に上記収容凹部3a全体を位置させる。
下方カメラ38が収容凹部3aを撮影する際、収容プレート3の上方からは上記パネル照明37が光を照射し、透明な収容プレート3の下方から収容された上記細胞塊1を明瞭に認識することが可能となっており、制御手段21は撮影された細胞塊1について、収容凹部3a内における位置を認識するとともに大きさ(外径寸法)と形状を計測する。
図7の各図は下方カメラ38が撮影した細胞塊1の撮影結果を示したものであり、このうち(a)に示す細胞塊1は形状が適切でなく、(b)に示す細胞塊1はその外径寸法が所定の大きさに達していない。
制御手段21は、このような細胞塊1については細胞の立体構造体1Aには使用しないものと判定し、吸引ノズル19による吸着保持動作を行わない。
一方、図7(c)(d)は形状ならびに外径寸法が適切である細胞塊1であり、(c)は細胞塊1が収容凹部3aの中央に位置し、(d)は細胞塊1が収容凹部3aの中央よりずれている場合を示している。
上記収容プレート3の収容凹部3aは底面が半球状であるため、(c)のように中央に位置している場合は、その細胞塊1は収容凹部3aにおける最も深い位置に位置し、また(d)のように中央からずれている場合は、その細胞塊1は収容凹部3aにおける最も深い位置よりも浅い位置に位置している。
制御手段21には、収容凹部3aの底面上の位置に対応する深さが把握されている。また、細胞塊1は略球状であるため、外径寸法(直径)からその高さを推定することができる。制御手段21は、このような収容凹部3a内の位置を認識するとともに、細胞塊1の外径寸法を計測することによって、細胞塊1を吸着保持する際における吸引ノズル19の下降量を設定する。
そして細胞塊1の外径は上記吸着部19aの内径よりも大径となっているため、細胞塊1は吸着部19aの内部に吸引されることなく先端に吸着保持されるようになっている。
このようにして吸引ノズル19に細胞塊1を吸着保持した後、上記吸引ノズル19は昇降手段36によって上昇され、上記パネル照明37が再び上記収容プレート3の上方に位置する。
上記下方カメラ38は今回の収容凹部3aを再び撮影し、細胞塊1が取り除かれたことを確認する。仮に細胞塊1が残っている場合には再度上記吸引ノズル19によって細胞塊1を吸着保持する工程をリトライする。
このとき上記リング照明25は収容容器7の上方に位置して支持体4を照明し、上方カメラ39によって針状体6の先端位置が認識されると旋回手段26によって退避される。
針状体6の先端位置が認識されると、制御手段21はX-Yユニット34を作動させて、図5(a)に示すように、細胞塊1を吸着保持した上記吸引ノズル19の吸着部19aを、支持体4における上記認識した針状体6の先端位置の上方まで移動させ、さらに吸引ノズル19を下降させて細胞塊1を針状体6の先端に突き刺す。
この状態から、図5(b)に示すように昇降手段36が吸引ノズル19を下方に移動させると、吸着部19aに上方から押圧された細胞塊1は針状体6によって貫通され、針状体6は上記吸着部19aの内部に挿入されていく。
また上記吸引ノズル19の下降量は事前に設定されているが、先に下方カメラ38で認識した細胞塊1の外径寸法に基づいて補正されるようになっている。
ここで、本実施例における上記吸着ノズル19の吸着部19aの内径を細胞塊1よりも小径にかつ針状体6の外周径(太さ)よりも大径とし、上記移動手段20は細胞塊1を吸着保持した吸引ノズル19を針状体6の軸方向に移動させることにより、細胞塊1を確実に針状体6に突き刺すことが可能となっている。
一方、針状体6の先端位置を認識した結果、細胞塊1を突き刺すべき針状体6の先端位置が規定範囲を外れている場合には、制御手段21は、図5(c)に示すように該外れている針状体6の先端位置に細胞塊1を突き刺し、その後、吸引ノズル19を水平方向に移動させ、先端位置を上記規定範囲内へと移動させ、これにより針状体6の姿勢を矯正させてから、吸引ノズル19を下降させる。
この場合、制御手段21は上記移送手段17を制御して、上記第3支持手段16に支持されている収容プレート3を上記第1支持手段14へと移送し、さらに移送手段17は上記第2支持手段15に支持された異種収容プレート3bを昇降テーブル24に載置させる。
その状態で、第2支持手段15の係合爪15aを退没させ、昇降テーブル24を下降させて収容プレート3bを上記第3支持手段16へと移送する。この状態で収容プレート3に収容された細胞塊1の場合と同様にして、異種収容プレート3bの収容凹部3aに収容された異種細胞塊1bについて処理を行う。
また、収容プレート3におけるすべての収容凹部3aから細胞塊1を取り出した場合、上記移送手段17は上記第3支持手段16に支持されている空の収容プレート3を回収マガジン13の下方へと移送し、回収マガジン13に収納する。

Claims (3)
- 1つの細胞塊を収容する収容凹部が多数形成された収容プレートと、上記細胞塊を突き刺して貫通する針状体を複数本備えた支持体と、負圧発生手段に接続されて上記細胞塊を吸着保持する吸引ノズルと、該吸引ノズルを上記収容プレートと支持体との間で移動かつ昇降させる移動手段と、上記吸引ノズルの吸引動作および上記移動手段の動作を制御する制御手段とを備え、
上記吸引ノズルは、先端に1つの細胞塊を吸着する管状の吸着部を備えるとともに、該吸着部の内径を、上記細胞塊の外径より小径に、かつ上記支持体の針状体の外周径よりも大径とし、
上記制御手段は、上記収容プレートの収容凹部において上記吸引ノズルの吸着部に上記細胞塊を吸着保持させると、上記移動手段によって該吸引ノズルを上記支持体における所要の針状体の上方に移動させ、
さらに該吸引ノズルを針状体の軸方向に移動させて上記細胞塊に針状体を突き刺し、上記細胞塊を貫通した針状体を上記吸着部の内部に挿入させることを特徴とする細胞の立体構造体製造装置。 - 上記収容プレートの収容凹部内の底面は中央が最も深い球面状に形成され、この収容凹部に収容された細胞塊を撮影する細胞塊撮影手段を備え、
上記制御手段は、該細胞塊撮影手段の撮影結果に基づいて、上記細胞塊の位置と外径とを認識して、上記移動手段によって上記吸着ノズルを細胞塊の上方に移動させるとともに、上記吸着部の先端を細胞塊に向けて接近させることを特徴とする請求項1に記載の細胞の立体構造体製造装置。 - 上記支持体の針状体を先端側から撮影する針状体撮影手段を備え、
上記制御手段は、該針状体撮影手段の撮影結果に基づいて、上記細胞塊を吸着保持した吸引ノズルを上記針状体の上方に移動させ、
上記針状体撮影手段によって撮影した針状体の先端位置が、所定の規定範囲内に位置していない場合、
上記制御手段は、上記吸引ノズルに吸着保持した細胞塊を針状体の先端に突き刺した状態で、上記吸引ノズルを移動させ、これにより針状体の先端位置を上記規定範囲内に位置させることを特徴とする請求項1または請求項2に記載の細胞の立体構造体製造装置。
